Target Information

Sclerostin은 주로 osteocyte에서 분비되는 당단백질로, Cerberus/DAN 계열의 BMP 길항 단백질에 속하며 Wnt 신호 경로의 내인성 조절자 역할을 합니다. 주로 뼈, 골수, 연골, 신장, 간 등에서 발현되며, hypertrophic chondrocyte 및 cementocyte에서도 생성됩니다.
Sclerostin은 LRP5 및 LRP6과 복합체를 형성하여 Wnt/β-catenin 신호를 억제하며, LRP4와의 상호작용을 통해 Wnt 신호 억제를 촉진합니다. 또한 osteocyte의 RANKL 발현을 증가시켜 골흡수 활성을 촉진할 수 있습니다.
이 단백질의 결핍은 sclerosteosis 및 van Buchem disease(VBD)와 같은 골 형성 이상 질환과 관련이 있습니다.
