Product Specific Information

The 10C4 monoclonal antibody reacts with mouse and rat Bcl-2, also known as B cell lymphoma 2. Bcl-2 is the founding member of the Bcl-2 family of proteins that regulate cell death and survival.
Members include pro-apoptotic proteins (BAD, Bax, Bim) and anti-apoptotic proteins (Bcl-2, Bcl-xL, Mcl-1). Bcl-2 inhibits apoptosis by preventing cytochrome c release from mitochondria.
Overexpression or mutation of Bcl-2 is implicated in various cancers. In thymocytes, Bcl-2 is highly expressed in CD4-CD8- double negative and CD4+/CD8+ single positive cells, but reduced in CD4+CD8+ double positive thymocytes.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated and tested by intracellular staining and flow cytometry of mouse splenocytes and thymocytes.
Use at 5 µL (1.0 µg) per test (final volume 100 µL, 10⁵–10⁸ cells/test).

eFluor® 450 is an alternative to Pacific Blue®. Excitation 405 nm, Emission 445 nm, detected with Violet laser.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

BCL-2 is a key regulator of apoptosis that can inhibit or promote cell death.
It forms dimers with other BCL-2 family members to modulate apoptosis.
Bag1 enhances BCL-2’s anti-apoptotic effect, while Bax and Bak promote cytochrome c release and apoptosis.
Constitutive BCL-2 expression, such as translocation to Ig heavy chain locus, is associated with follicular lymphoma.
Overexpression of BCL-2 has been linked to cancers including B-cell lymphoma and prostate cancer.
Two transcript variants exist, differing at the C-terminal end.
