Target Information
LSD1 is a histone demethylase that specifically demethylates Lys-4 of histone H3, a marker for epigenetic transcriptional activation, thereby acting as a corepressor. LSD1 contains a SWIRM domain, a FAD-binding motif, and an amine oxidase domain. It is part of several histone deacetylase complexes and silences genes by functioning as a histone demethylase. The enzyme oxidizes the substrate via FAD to generate an imine, which is subsequently hydrolyzed. LSD1 demethylates both mono- and tri-methylated Lys-4 of histone H3 and may play a role in the repression of neuronal genes. It requires RCOR1/CoREST to demethylate H3 Lys-4 on nucleosomes. LSD1 may also demethylate Lys-9 of histone H3, leading to derepression of androgen receptor target genes. Mutations in this gene can cause cleft palate, psychomotor retardation, and distinctive facial features.
