Product Specific Information
The O323 monoclonal antibody reacts with human CD27, a lymphocyte-specific member of the TNFR superfamily. CD27 is expressed by a subset of thymocytes and virtually all mature T cells and is upregulated upon T-cell stimulation. CD27 binds to CD70, playing an important role in T cell–B cell interaction.

- Applications Tested: Pre-diluted and tested on normal human peripheral blood cells
- Use at 5 µL (0.25 µg) per test
- A test = staining a cell sample in 100 µL final volume
- Recommended cell count: 10⁵–10⁸ cells/test
Super Bright 702 is a tandem dye excited with a violet laser (405 nm) and emits at 702 nm. Recommended filter: 710/50 bandpass.
Use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.
Light-sensitive: Protect vial and stained samples from light.
Fixation:
Samples can be stored up to 3 days in the dark at 4°C using:
- IC Fixation Buffer (Product # 00-8222)

Target Information
CD27 is a 50 kDa member of the TNF receptor superfamily involved in regulation of cell proliferation and death. Expression of CD27 and its ligand CD70 is mainly confined to lymphocytes. CD27 forms disulfide-linked homodimers on mature thymocytes, peripheral blood T cells, and a subset of B cells. Activation via TCR-CD3 complex increases CD27 expression and release of soluble CD27 (sCD27), a prognostic marker in B cell malignancies.
CD27-binding protein (SIVA) is a proapoptotic protein involved in CD27-mediated apoptosis.
Diseases associated with CD27 dysfunction include Lymphoproliferative Syndrome 2 and Autosomal Recessive Lymphoproliferative Syndrome.
