Target Information
The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or pyrimidine nucleotide starvation. Upon stress, p53 activates transcription of genes such as p21waf1, leading to G1 or G2/M arrest.
Structurally, p53 includes:
- N-terminal transactivation domain
- Central DNA-binding domain
- Oligomerization domain
- C-terminal regulatory domain
Phosphorylation at Ser15 is critical for activation and stabilization. p53 blocks proliferation of damaged cells and acts as an anticancer agent. Phosphorylation at Ser392 is associated with tumor formation. It is also linked to aging, oxidative stress, and impaired learning due to reduced LTP.
Normal cells express p53 at low levels, while transformed cell lines show high expression, contributing to malignancy. Mutant p53 loses DNA binding and tumor suppressor activity. TP53 gene alterations occur in somatic mutations in cancers and germline mutations in Li-Fraumeni syndrome.
