Description:
The M17/4 monoclonal antibody reacts with mouse CD11a, the 180 kDa integrin alpha L (LFA-1α). LFA-1, formed by non-covalent association of CD11a with CD18 (integrin β2), is an adhesion molecule involved in lymphocyte and granulocyte function. Ligands include CD54 (ICAM-1), CD102 (ICAM-2), and CD50 (ICAM-3). CD11a is expressed by all leukocytes.

• Blocking of CD11a function

Applications Tested:
Tested by flow cytometry using mouse splenocytes. Recommended ≤0.5 µg/test in 100 µL final volume. Cell number: 10⁵–10⁸ cells/test. Titrate for optimal performance.

Storage and Handling:
Use in a sterile environment. Store at 4°C.

Filtration:
0.2 µm post-manufacturing filtered.

Target Information

CD11a (LFA-1α, ITGAL) forms part of the LFA-1 complex with CD18 (αLβ2 integrin). It mediates leukocyte adhesion via ICAM-1, -2, and -3, and participates in lymphocyte costimulatory signaling. CD11a expression is found on leukocyte membranes, shifting from low- to high-affinity conformations upon stimulation. It is critical in immune cell interaction, migration, and T-cell function regulation.

Blocking CD11a is a therapeutic strategy for inflammatory diseases. Humanized anti-CD11a antibody Efalizumab (Raptiva) and CD11a-binding compounds like simvastatin demonstrate immunomodulatory effects. Two transcript variants of CD11a are known. Diseases linked to CD11a dysfunction include benign cephalic histiocytosis and leukocyte adhesion deficiency.
