Product Specific Information

Description:
The 53-6.7 monoclonal antibody reacts with the mouse CD8a molecule, a 32–34 kDa cell surface receptor expressed as either CD8αβ heterodimer or CD8αα homodimer. CD8 binds MHC class I and, via p56lck association, plays a key role in T cell development and activation.

Applications Reported:
Useful for flow cytometric analysis, T cell activation, isolation and depletion of CD8+ cells, and blocking of ligand binding.

Applications Tested:
Validated by flow cytometric analysis of mouse thymocytes and splenocytes. Recommended ≤0.25 µg/test (100 µL final volume, 10^5–10^8 cells/test). Optimize titration for best assay performance.

Target Information

Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein and co-receptor for MHC class I molecules. It is primarily expressed on T cells as CD8αβ heterodimers and on subsets of T cells, NK cells, monocytes, and dendritic cells as CD8αα homodimers. CD8 engagement with MHC-I/peptide complexes recruits Lck kinase, leading to CTL activation and immune response. Differentiation of CD8+ T cells into cytotoxic T lymphocytes is enhanced by IL-2, IL-12, and TGF-β1.
