Product Specific Information

The CBRM1/5 monoclonal antibody binds an activation-specific epitope of human Mac-1. It recognizes Mac-1 molecules on neutrophils and monocytes after stimulation but not on resting myeloid cells. Mac-1 mediates adhesive cell interactions, and the epitope recognized by CBRM1/5 is located near the ligand-binding site on the alpha chain I domain.

CBRM1/5 blocks Mac-1–dependent adhesion to fibrinogen and ICAM-1 and inhibits chemoattractant-stimulated adhesion of eosinophils to ICAM-1. Some binding may occur to Mac-1 in unstimulated blood samples due to low-level activation.

Target Information

CD11b (integrin alpha-M, ITGAM, integrin alpha-X, ITGAX) is a 165 kDa adhesion molecule that forms a heterodimer with CD18 (integrin beta-2). The CD11b/CD18 complex (Mac-1, CR3) is expressed on monocytes/macrophages, granulocytes, activated lymphocytes, subsets of NK and dendritic cells, and microglia.

It functions as a receptor for ICAM-1, ICAM-2, ICAM-4, CD14, CD50, CD23, heparin, iC3b, fibrinogen, and Factor X, playing a key role in cell-cell and cell-matrix interactions. Expression varies by cell type and activation state. CD11b is used as a microglial marker and is linked to conditions such as systemic lupus erythematosus and ITGAM-related susceptibility to autoimmune disease.
