Product Specific Information

The eBioL31 monoclonal antibody reacts with mouse CD207 (Langerin), expressed in Langerhans cells (LC), a subset of dendritic cells. Mouse CD207 is a 48 kDa C-type lectin transmembrane protein involved in antigen recognition and uptake. LC in the epidermis reduce CD207 expression upon activation and migrate toward lymphatic vessels. Expression correlates with Birbeck Granules formation, also observed in fibroblast cell lines transfected with CD207 cDNA.

Western blotting with eBioL31 shows a 48 kDa band in mouse ear epidermis lysate and CD207-transfected cells. Preliminary data suggest eBioL31 can be used for intracellular flow cytometry with the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523). The recognized epitope is in the extracellular domain within the CRD, enabling detection of both surface and intracellular langerin. Expression differences may occur between mouse strains.

Applications Tested: Flow cytometric analysis of mouse CD207-transfected NRK cells (≤0.06 µg/test).

Target Information

Langerin (CD207) is a type II membrane-associated C-type lectin expressed exclusively by Langerhans cells. It recognizes mannose residues via its carbohydrate recognition domain (CRD) and is localized both on the cell surface and intracellularly near Birbeck granules. Transfection of Langerin cDNA into fibroblasts induces formation of Birbeck granules, demonstrating its role in membrane superimposition and BG formation.
