Target Information
Caspase 9 (ICE-like apoptotic protease 6, ICE LAP6, apoptotic protease Mch6, apoptotic protease activating factor 3, Apaf3)는 CARD 도메인을 포함한 peptidase family C14의 구성원입니다. Caspases는 시스테인 프로테아제로, 비활성 전구체 형태로 발현되며 특정 기질의 절단을 통해 세포사멸(apoptosis)을 매개합니다.
Caspase 9는 heterotetramer 형태로 활성화되며, 세포질에 존재하다가 활성화 시 미토콘드리아로 이동합니다. Apaf-1과 결합하여 caspase-3 및 caspase-6을 활성화시키는 세포사멸 경로의 핵심 효소입니다. 또한 DNA 손상에 의한 apoptosis를 ABL1/c-Abl 의존적으로 촉진하고, PARP(poly(ADP-ribose) polymerase)를 절단합니다.
Caspase 9는 다양한 인체 조직에서 발현되며, 결핍 시 뇌 발달 이상을 유발할 수 있습니다. 비정상적인 Caspase 9 발현은 암 및 알츠하이머병(AD)과 같은 신경퇴행성 질환과 관련이 있습니다.
