Product Specific Information

The B-Ly7 monoclonal antibody reacts with human CD103, the alpha E integrin. CD103 non-covalently associates with integrin beta 7 and is mainly expressed on intraepithelial lymphocytes and a small subset of peripheral lymphocytes. It is also expressed by hairy cell leukemia (HCL) and some chronic B cell lymphocytic leukemias. In vitro stimulation of human T cells with mitogens induces upregulation of CD103. E-cadherin binds to CD103 and mediates homing of lymphocytes to the intestinal epithelium.

Each product contains:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested by flow cytometric analysis of 2–3 day PHA-activated human peripheral blood cells. Recommended use: 5 µL (0.4 µg) per test (100 µL final volume).

Note: NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

This conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which provides improved staining performance. Add 5 µL per sample to antibody cocktails prior to combining with cells. For single-color controls, use 5 µL per 100 µL cell sample.

NovaFluor conjugates utilize Phiton™ technology with engineered fluorescent signatures to minimize spillover and spread.

Target Information

CD103 (ITGAE, Integrin alpha E) is expressed on mucosa-associated T lymphocytes and activated cells. It consists of a 150 kDa alpha chain and a 120 kDa beta chain. CD103 is expressed by 0.5–2% of resting lymphocytes in peripheral blood and lymphoid organs, and stains a few bone marrow cells. It forms a complex with beta 7 integrin to bind E-cadherin, mediating adhesion and activation of intestinal intraepithelial lymphocytes (IEL).
Diseases associated with CD103 dysfunction include hairy cell leukemia and splenic marginal zone lymphoma.
