Product Specific Information

The monoclonal antibody IgE21 recognizes the immunoglobulin E (IgE) class of antibodies. IgE levels in serum are low and play a key role in allergic responses by binding to mast cells and basophils through the Fc epsilon receptor I, leading to histamine release. IgE is produced by terminally differentiated plasma B cells and basophils and exists in both soluble and membrane-bound forms. IgE21 recognizes both.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells at 5 µL (0.2 µg) per test.
A test is defined as the amount of antibody needed to stain a cell sample in 100 µL final volume (10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining.
• Add 5 µL of CellBlox Plus per sample, regardless of the number of NovaFluors used.
• For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates are based on Phiton™ technology, utilizing novel nucleic acid dye structures for engineered fluorescent signatures optimized for minimal spillover and spread.

Target Information

IgE is one of five immunoglobulin classes in mammals. It consists of two heavy (epsilon) chains and two light chains. IgE mediates type I hypersensitivity reactions (e.g., hay fever, asthma, anaphylaxis) by binding to Fc receptors on basophils and mast cells, triggering degranulation. It is also important for defense against parasitic infections.
