Product Specific Information

The monoclonal antibody IgE21 recognizes the immunoglobulin E (IgE) class of antibodies. Natural IgE levels are low in serum and play a key role in allergy responses by binding to mast cells and basophils via the Fc epsilon receptor I, triggering histamine release. IgE is produced by terminally differentiated plasma B cells and basophils, and exists in soluble and membrane-bound forms—both recognized by IgE21.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells at 5 µL (0.2 µg) per test. A test is defined as staining a cell sample in 100 µL volume (10⁵–10⁸ cells/test).

NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.

This conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which improves formulation over the previous CellBlox version.
Use 5 µL per sample for optimal staining, regardless of the number of NovaFluors in the panel. For single-color controls, add 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, based on novel nucleic acid dye structures engineered for optimized fluorescence signatures and reduced spectral spillover.

Target Information

IgE is one of five immunoglobulin classes in mammals, consisting of two epsilon heavy chains and two light chains. It plays a critical role in type I hypersensitivity reactions (e.g., hay fever, asthma, hives, anaphylactic shock) by binding to Fc receptors on basophils and mast cells. Crosslinking of receptor-bound IgE triggers degranulation and allergic responses. IgE also contributes to parasite immunity.
