Target Information

STAT4 was originally identified using degenerate primers complementary to conserved regions of other STAT proteins. The STAT4 protein shares 52% similarity with STAT1 and 47% with STAT3. It can be tyrosine phosphorylated by Jak1 or Jak2, forming homo- and heterodimers with related STAT family members.

Tyrosine-phosphorylated STAT4 binds the IFN-γ activated site (GAS), and serine phosphorylation is required for maximal transcriptional activity. STAT4 expression is mainly found in the thymus, spleen, and testis.

STAT4 is activated by interleukin 12 (IL-12), which is critical for T-cell–independent induction of IFN-γ and the Th1 immune response against intracellular pathogens. STAT4-deficient mice show impaired IL-12–induced Th1 development and enhanced Th2 differentiation.

Recent studies indicate that IFN-α/β activation of STAT4 is required for IFN-γ production during viral infection.
