Target Information

TAK1 (also known as MAP3K7) is a serine/threonine kinase involved in multiple signaling pathways. Initially identified as a TGF-β activated kinase, TAK1 interacts with TGF-β receptors and TRAF6 to modulate activation of JNK and p38. It also participates in Wnt, BMP, and activin signaling, and responds to thyroid hormone, osmotic stress, and endothelin. TAK1 regulates wound healing via activation of von Hippel-Lindau tumor suppressor and represses PDGF-B and integrins. Furthermore, TAK1 is essential for IKK activation through IL-1, IL-6, IL-18, TNF, CD40, TLR, and RIG-I pathways. Activation involves TAB2/3 binding, polyubiquitination, and TAK1 autophosphorylation at Thr178, Thr184, Thr187, and Ser192, leading to phosphorylation of IKK-β. TAK1 also represses human telomerase reverse transcriptase, influencing cell lifespan regulation.
