Target Information
IGF1R (Insulin-like Growth Factor 1 Receptor) is a receptor tyrosine kinase activated by IGF-1 and IGF-2. It plays a key role in cell growth and survival and is implicated in various cancers. The receptor is a tetramer composed of two alpha and two beta subunits. The alpha subunits bind IGF-1 with higher affinity than IGF-2, triggering autophosphorylation at Tyr1134, Tyr1135, and Tyr1136. Activated IGF1R subsequently phosphorylates substrate proteins, leading to activation of RAS, RAF, and MAPK pathways (ERK, p38, JNK), which promote gene transcription and proliferation.
