Target Information
X-linked ectodysplasin-A2 receptor (XEDAR) is a member of the tumor necrosis factor receptor family, highly expressed during embryonic development and binds to ectodysplasin-A2 (EDA-A2). Two main isoforms, XEDAR-s and XEDAR-L, differ by a 21-amino acid region at the juxtamembrane region of the cytoplasmic domain. Both lack a death domain and signal mainly through TRAF3 and TRAF6 to activate NF-κB and JNK pathways. EDA-A2 treatment of XEDAR-transfected cells induces a complex containing FADD, caspase-8, and caspase-10, leading to activation of caspase-8, caspase-3, and apoptosis. This apoptosis is caspase-9 dependent and can be blocked by inhibitors of caspase-8.
