• Preparation:
  Produced from sera of rabbits immunized with highly pure Recombinant Human MMP-2. Anti-Human MMP-2-specific antibody was purified by affinity chromatography employing an immobilized Human MMP-2 matrix.

• Sandwich ELISA:
  To detect Human MMP-2 by sandwich ELISA (using 100 µL/well), use 0.5–2.0 µg/mL of this antibody. When paired with PeproTech Biotinylated Anti-Human MMP-2 (500-P307BT) as detection antibody, the assay detects at least 2000–4000 pg/mL of Recombinant Human MMP-2.

• Western Blot:
  For Human MMP-2 detection by Western blot, use 0.1–0.2 µg/mL. Detection limit for Recombinant Human MMP-2 is 1.5–3.0 ng/lane under reducing or non-reducing conditions.

Target Information

Matrix metalloproteinases (MMPs) are proteolytic enzymes capable of degrading connective tissue components. They share a conserved activation mechanism and metal-binding active site and are inhibited by tissue inhibitors of metalloproteinases (TIMPs). MMPs and TIMPs regulate angiogenesis and tissue remodeling.

MMP2 is synthesized as a 631 amino acid proenzyme activated by cleavage of the first 80 amino acids, with propeptide, catalytic, and hemopexin domains. MMP2 mutations are associated with Winchester syndrome and Nodulosis-Arthropathy-Osteolysis (NAO) syndrome. Two transcript variants encoding different isoforms of MMP2 have been identified.
