Product Specific Information
This antibody binds CD33, a 67 kD glycoprotein expressed on monocytes, hematopoietic progenitors, early myeloid progenitor cells, and myeloid leukemias (acute and chronic). It is not expressed on mature myeloid cells, normal stem cells, other hematopoietic cells, or non-hematopoietic human tissues.
Target Information
CD33 is a transmembrane protein belonging to the sialic acid-binding immunoglobulin-like lectin (Siglec) family. It contains immunoreceptor tyrosine-based inhibitory motifs (ITIMs) that recruit SHP-1 and SHP-2 phosphatases and participate in receptor regulation. CD33 is expressed on cells of the myelomonocytic lineage, binds sialic acid residues on cell surfaces, and serves as a therapeutic target for acute myeloid leukemia. It is found on granulocyte and macrophage precursors in bone marrow, but not on pluripotent stem cells. CD33 is also expressed on peripheral monocytes and is useful for distinguishing myelogenous leukemia cells from lymphoid or erythroid leukemias. Diseases associated with CD33 dysfunction include gallbladder lymphoma and extracutaneous mastocytoma.
