Target Information
Epidermal Growth Factor (EGF) binds to the EGF receptor (EGFR), a 170 kDa protein kinase, initiating diverse cellular signaling pathways. EGFR forms homo- or heterodimers with other family members, each activating distinct signaling cascades through SH2-containing effector proteins. EGF is expressed in kidney, cerebrum, prostate, and salivary glands, acting as a potent mitogenic factor. Phosphorylated EGFR recruits adapter proteins such as GRB2 to activate downstream pathways including RAS-RAF-MEK-ERK, PI3K-AKT, PLCγ-PKC, and STAT. Research suggests EGF also plays a role in NF-κB signaling. Defects in the EGF gene are linked to hypomagnesemia type 4 and may contribute to cancer progression.
