Product Specific Information

The RPA-T4 monoclonal antibody reacts with human CD4, a 59 kDa cell surface receptor expressed by most thymocytes, a subset of mature T-helper cells, and at low levels on monocytes. CD4 serves as a receptor for HIV, and RPA-T4 blocks HIV binding and mixed lymphocyte reactions. It recognizes a different epitope than the OKT4 antibody, and the two do not cross-block each other.

• Applications Reported: Flow cytometric analysis
• Applications Tested: Pre-diluted and validated for flow cytometry using normal human peripheral blood cells. Use 5 µL (0.125 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).
• Fluorochrome: Brilliant Ultra Violet™ 661 (BUV661) — emits at 660 nm, for use with 355 nm UV laser cytometers.
• Buffer Recommendation: When using multiple polymer dyes, use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific interactions.
• Light Sensitivity: Protect from light due to photo-induced oxidation.
• Fixation: Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C in the dark with minimal signal loss.

Target Information

CD4 is a co-receptor for MHC class II molecules and HIV. It is primarily expressed on T-helper cells, and also on monocytes, macrophages, and dendritic cells. CD4 expression is found in thymus, lymph nodes, tonsils, spleen, and certain non-lymphoid tissues such as brain and gut. It assists in T-cell activation by associating with the T-cell receptor complex and protein tyrosine kinase Lck, and may mediate neuronal damage in immune-related CNS diseases. Multiple alternatively spliced transcripts have been identified (RefSeq, July 2017).
