Product Specific Information

The GK1.5 monoclonal antibody reacts with mouse CD4, a 55 kDa cell surface receptor expressed on thymocytes, subsets of mature T cells, and dendritic cells. CD4 binds to MHC class II molecules on antigen-presenting cells and is essential for T-cell development and function. In T cells, CD4 associates with protein tyrosine kinase p56lck via its cytoplasmic domain. Binding of GK1.5 is blocked by RM4-5.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes at ≤ 0.2 µg/test (in 100 µL final volume, 10⁵–10⁸ cells/test). Titrate for optimal performance.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell Stains for viability assessment.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample (regardless of the number of NovaFluors).
  • For single-color controls, add 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Target Information

CD4 is a co-receptor for MHC class II molecules and plays a key role in T-cell activation and immune signaling. It is primarily expressed on T-helper cells but also found on monocytes, macrophages, and dendritic cells. CD4 expression is seen in thymus, lymph nodes, tonsils, spleen, and certain non-lymphoid tissues. It associates with Lck kinase to mediate T-cell receptor signaling and may contribute to neuronal damage in immune-mediated CNS diseases.
