Product Specific Information

Description:
The GK1.5 monoclonal antibody reacts with the mouse CD4 molecule (55 kDa), expressed by most thymocytes, subsets of mature T cells, and dendritic cells. CD4 binds to MHC class II molecules on antigen-presenting cells and is essential for T cell development and function. In T cells, CD4 associates with protein tyrosine kinase p56lck. Binding of GK1.5 is blocked by RM4-5.

Each product includes one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

Applications Reported:
Validated for flow cytometric analysis.

Applications Tested:
Tested by flow cytometry using mouse splenocytes at ≤ 0.2 µg/test. A test is defined as the amount of antibody staining a cell sample in 100 µL final volume. Recommended to titrate for optimal performance.

Compatibility Notes:
NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, and DAPI. Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for best results.

Blocking Buffer Guidance:
This conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which enhances staining performance. Add 5 µL per sample to antibody cocktails before combining with cells. For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Target Information

CD4 Function:
CD4 is a co-receptor involved in MHC class II recognition and T-cell activation. It is primarily expressed on T helper cells, as well as monocytes, macrophages, and dendritic cells. CD4 expression is found in thymus, lymph nodes, tonsils, spleen, and certain non-lymphoid tissues. It interacts with the T-cell receptor complex and protein tyrosine kinase Lck, playing a key role in immune signaling and potentially in neuronal damage during immune-mediated diseases.
