Product Specific Information
The GK1.5 monoclonal antibody reacts with the mouse CD4 molecule, a 55 kDa cell surface receptor expressed by most thymocytes, subsets of mature T cells, and dendritic cells. CD4 binds MHC class II on antigen-presenting cells and plays a critical role in T cell development and function. In T cells, CD4 associates with protein tyrosine kinase p56^lck through its cytoplasmic tail. Binding of GK1.5 is blocked by RM4-5.

Applications Reported: Flow cytometric analysis
Applications Tested: Mouse splenocytes, ≤0.2 µg/test (100 µL final volume, 10⁵–10⁸ cells/test)
Usage Note:
- Carefully titrate for optimal performance.
- NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
- Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
- CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
- Add 5 µL per sample before combining with cells.
- For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells).

Target Information
CD4 is a co-receptor involved in the recognition of MHC class II molecules and HIV binding. It is mainly expressed in T helper cells but also in monocytes, macrophages, and dendritic cells. Tissue expression includes thymus, lymph nodes, tonsils, spleen, and certain regions of brain and gut. CD4 participates in early T-cell activation via association with the T-cell receptor complex and Lck kinase, and may mediate neuronal damage in immune-related CNS diseases. Multiple alternatively spliced transcripts have been identified.
