Target Information
ERN1 serine/threonine kinase, also known as endoplasmic reticulum to nucleus signaling 1 (IRE1), is a human homologue of the yeast Ire1 gene product. It possesses intrinsic kinase and endoribonuclease activities that play a critical role in endoplasmic reticulum (ER)-based stress signaling.
ERN1 regulates IRE1 proteolysis in mammalian cells and is essential for fetal viability through its function in extraembryonic cells.
IRE1 acts as a sensor of unfolded proteins in the ER, initiating the unfolded protein response (UPR) by splicing XBP1 mRNA into a potent transcriptional activator. While null mutants are viable under normal conditions, IRE1 is essential under stress conditions that lead to unfolded protein accumulation in the ER.
Depletion of IRE1p or expression of a dominant negative form has minimal effect under normal conditions but induces apoptosis under ER stress.
