Target Information
KLOTHO is a systemic anti-aging hormone belonging to the glycosidase1 superfamily. It encodes a type I membrane protein highly expressed in the kidney and brain. In mice, KLOTHO deficiency leads to aging-like phenotypes such as arteriosclerosis, osteoporosis, and skin atrophy, along with growth retardation and infertility.
Overexpression of KLOTHO extends lifespan by inhibiting insulin/IGF1 signaling. It also regulates calcium/phosphorus homeostasis by inhibiting active vitamin D synthesis and is identified as a potential tumor suppressor.
