Description:
The M18/2 monoclonal antibody reacts with mouse CD18, a 90–95 kDa integrin beta 2 glycoprotein. CD18 is the common beta subunit that associates with CD11a (alpha L), CD11b (alpha M), and CD11c (alpha X) to form LFA-1, Mac-1, and gp150/95 complexes. These heterodimers are broadly expressed by leukocytes, with NK and T cells showing higher expression levels. CD18 is involved in cellular adhesion and immune response through interactions with its counter-receptors.

Applications Reported:
Reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes and bone marrow cells. Recommended use ≤ 0.5 µg per test (in 100 µL final volume). Cell number may range from 10⁵–10⁸ cells/test. Titrate antibody for optimal performance.

0.2 µm post-manufacturing filtered.

Target Information

CD18 (integrin beta 2 subunit) is a 90 kDa type I transmembrane protein expressed on all leukocytes. It forms heterodimers with CD11a–d molecules to constitute leukocyte (beta2) integrins:

• αLβ2 (CD11a/CD18, LFA-1)
• αMβ2 (CD11b/CD18, Mac-1, CR3)
• αXβ2 (CD11c/CD18)
• αDβ2 (CD11d/CD18)

These integrins mediate cell adhesion and signaling critical for leukocyte migration. CD18 deficiency causes leukocyte adhesion deficiency type I (LAD1), and reduced expression can lead to psoriasiform skin disease. CD18 also mediates leukotoxin-induced cytolysis by Mannheimia haemolytica.
