Target Information

VEGF (Vascular Endothelial Growth Factor)는 45 kDa의 이황화결합된 호모다이머 글라이코프로틴으로, 혈관신생(angiogenesis)에 관여하며 종양 성장과 전이를 촉진합니다.
VEGF는 내피세포의 생존, 유사분열, 화학주성, 혈관 투과성 증가에 영향을 미칩니다. VEGF 패밀리에는 VEGF-A, VEGF-B, VEGF-C, VEGF-D, VEGF-E, PlGF가 포함됩니다.
VEGF와 그 수용체(VEGFR-1, VEGFR-2)는 혈관신생의 세포 신호 조절의 핵심 요소로 알려져 있습니다.
대부분의 종양은 혈관신생이 필수적이며, VEGF는 이 과정에서 가장 강력한 혈관신생 유도 사이토카인으로 알려져 있습니다.
VEGF 유전자의 대체 스플라이싱으로 생성된 5가지 아이소폼이 존재하며, 이들은 주로 내피세포에 발현되는 티로신 키나아제 수용체 FLT1(FLT-1) 및 FLK1/KDR(FLT-2)에 결합합니다.
또한 FLT1의 대체 스플라이싱 형태인 sFLT1은 VEGF를 높은 친화도로 결합하여 혈관신생 억제 역할을 합니다.
VEGF의 과발현은 POEMS 증후군(Polyneuropathy, Organomegaly, Endocrinopathy, Monoclonal gammopathy, Skin changes)과 관련이 있습니다.
