Product Specific Information

The GIGD7 monoclonal antibody reacts with mouse TIGIT (T cell immunoreceptor with Ig and ITIM domains, also known as VSTM3 or WUCAM), a 26 kDa protein in the poliovirus receptor (PVR) family.

• Expression: In mice, TIGIT is found in follicular T helper cells; in humans, on NK cells, regulatory T cells, follicular T helper cells, memory CD4+ and CD8+ T cells.
• Not expressed on: B cells or naive CD4+ T cells.
• Function: TIGIT interacts with PVR and PVR-like proteins (e.g., CD155), mediating interactions with antigen-presenting cells, fibroblasts, and endothelial cells.

Applications Reported: Suitable for flow cytometric analysis (fluorochrome-conjugated GIGD7 recommended).

Applications Tested: Validated by flow cytometry using mouse TIGIT-transfected cell lines.
Use ≤0.25 µg/test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Optimize by titration for best performance.

Storage and Handling: Use under sterile conditions.
Filtration: 0.2 µm post-manufacturing filtered.
Purity: >90% (SDS-PAGE).
Endotoxin Level: <0.001 ng/µg antibody (LAL assay).
Aggregation: <10% (HPLC).

Target Information

TIGIT binds with high affinity to the poliovirus receptor (PVR), leading to increased IL10 and decreased IL12B secretion, and suppresses T cell activation by promoting mature immunoregulatory dendritic cell generation.
