Product Specific Information

The 3D12 monoclonal antibody reacts with human CCR7 (CD197), also known as EBI-1.
CCR7 is a G-protein-coupled chemokine receptor with seven transmembrane domains and functions as a receptor for CCL19 and CCL21. It is expressed on T cells and dendritic cells (DCs), distinguishing naïve, central, and effector memory T cell subsets. CCR7 expression increases during DC maturation and is involved in chemotaxis, endocytosis regulation, survival, and maturation.

• Recommended incubation: ≥45 minutes at 2–8°C for optimal staining.
• Applications Reported: Flow cytometric analysis.
• Applications Tested: Pre-diluted and validated for flow cytometry of human peripheral blood cells at 5 µL (1 µg)/test.
• Excitation/Emission: 405 nm / 600 nm (Violet laser).
• Recommended filter: 610/20 bandpass.
• Light sensitivity: Protect from light; dye is sensitive to photo-induced oxidation.
• Fixation: Samples can be stored for up to 3 days at 4°C in the dark using IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) with minimal effect on brightness and FRET efficiency.
• Buffer recommendation: When using multiple Super Bright dyes, use Super Bright Complete Staining Buffer (Product #SB-4401) to minimize polymer interactions.

Target Information

CCR7 is a G protein-coupled receptor in the chemokine receptor family, induced by Epstein-Barr Virus (EBV).
It is expressed in blood, bone marrow, lymph nodes, and intestinal tissues, particularly in activated B and T lymphocytes. CCR7 regulates memory T cell migration to inflamed tissues and dendritic cell maturation.
The specific ligands for CCR7 are CCL19 (ECL) and CCL21. Other receptors in the C-C chemokine family include CCR1, CCR2A, CCR3, CCR4, CCR5, and the Duffy blood group antigen, which play roles in T cell chemotaxis and phagocyte migration.
