Product Specific Information

Description:
The 205yekta antibody reacts with mouse CD205 (DEC-205), a 205 kDa integral membrane glycoprotein involved in antigen uptake, trafficking, and presentation that enhances antigen-specific T cell immunity. CD205 is highly expressed by CD8+ dendritic cells and at varying levels by bone marrow Gr1+ cells, Langerhans cells, bone marrow–derived dendritic cells (BMDCs), and thymic epithelial cells.

Important Note:
CD205 can bind to PE-Cy5.5 antibody and streptavidin conjugates; avoid using this format on CD205⁺ dendritic cells.

Applications Reported:
Validated for flow cytometric analysis.

Applications Tested:
Tested by flow cytometry on mouse bone marrow cells at ≤ 0.125 µg/test (100 µL final volume). Optimal antibody concentration should be titrated empirically. Typical cell count: 10⁵–10⁸ cells/test.

Fluorochrome Information:
Super Bright 600 is a tandem dye excited by the violet laser (405 nm) and emits at 600 nm. Recommended filter: 610/20 bandpass. Verify instrument compatibility.

Staining Buffer Recommendation:
When using multiple Super Bright dyes, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity:
Protect vial and stained samples from light to prevent photo-induced oxidation.

Fixation:
Samples may be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-57) for up to 3 days at 4°C in the dark with minimal loss of brightness or FRET efficiency.

Target Information

CD205 (LY25, Lymphocyte Antigen 75) is an endocytic membrane glycoprotein that facilitates antigen uptake and presentation, enhancing T cell immunity. It is highly expressed in CD8⁺ dendritic cells and variably in bone marrow Gr1⁺ cells, Langerhans cells, BMDCs, and thymic epithelial cells. CD205 directs captured antigens to antigen-processing regions and may reduce B lymphocyte proliferation.
Associated diseases include pneumonic plague and lipid pneumonia.
