Description:
The eBioC9B7W monoclonal antibody recognizes mouse CD223 (LAG-3, LAG3) protein expressed by activated α/β-TCR bearing T cells, a subset of γ/δ-TCR bearing T cells, and NK cells. CD223 is a 70 kDa type I transmembrane protein structurally similar to CD4. A soluble form can be generated by proteolytic cleavage, producing a 54 kDa extracellular fragment and a 16 kDa intracellular fragment.

CD223 binds to MHC class II with higher affinity than CD4 and is involved in negative regulation of T-cell activation and homeostatic proliferation. It is expressed by CD4+CD25+ regulatory T cells and may play a role in their regulatory function.

Applications Reported:
Reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometry using stimulated mouse splenocytes at ≤1.0 µg/test. Optimal titration is recommended. A test corresponds to staining a cell sample (100 µL) containing 10⁵–10⁸ cells.

Fluorochrome Characteristics:
Super Bright 600 is a tandem dye excited by the violet laser (405 nm) and emits at 600 nm. Recommended filter: 610/20 bandpass. Ensure instrument compatibility.

Staining Recommendations:
When using multiple Super Bright-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific interactions.

Light Sensitivity:
Protect from light to prevent photo-induced oxidation.

Fixation:
Samples may be fixed with IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal impact on brightness.

Target Information

LAG-3 is a 70 kDa surface glycoprotein in the Ig superfamily with homology to CD4. It binds MHC class II with higher affinity than CD4 and negatively regulates T cell activation and homeostasis. Expressed on activated T cells (including regulatory T cells) and NK cells, with higher expression on CD8+ T cells. Coexpression with CD49b identifies Type 1 regulatory T cells (Tr1).
