Target Information
p53는 세포 스트레스에 의해 활성화되는 종양 억제 단백질로, DNA 손상이나 뉴클레오타이드 결핍 시 세포 주기 정지 또는 세포사멸을 유도합니다. p53은 스트레스 신호에 반응하여 p21waf1 등의 유전자 발현을 촉진하며, G1 또는 G2/M 단계에서 세포 주기를 억제합니다.
구조적으로 N-말단 전사활성화 도메인, 중앙 DNA 결합 도메인, 올리고머화 도메인, C-말단 조절 도메인을 포함합니다.
Ser15, Ser392 등의 인산화 부위는 p53의 활성 및 안정화에 중요한 역할을 합니다. 특히 Ser392 인산화는 종양 형성과 관련이 있으며, 노화 및 산화 스트레스에도 연관되어 있습니다.
정상 세포에서는 낮은 수준으로 존재하지만, 다양한 변형 세포주에서는 높은 수준으로 발현되어 암화에 기여할 수 있습니다. TP53 유전자의 변이는 여러 인체 암에서 발견되며, Li-Fraumeni 증후군과 같은 유전적 암 소인 가족에서도 보고됩니다.
