Product Specific Information
PA1-072 detects munc-18 interacting protein 3 (Mint3) from mouse cells and rat brain samples.
It has been successfully used in Western blot procedures, detecting an ~86 kDa protein representing Mint3 from AtT20 cell extract and rat brain samples.
The immunogen is a synthetic peptide corresponding to residues M(1)EFLPEPQHPPGPPTMDLE(19) of rat Mint3.
The immunizing peptide (Cat. # PEP-135) is available for use in neutralization and control experiments.
Target Information
The munc-18 interacting protein (Mint) family comprises conserved adaptor proteins involved in membrane transport and organization.
In mammals, three isoforms exist: Mint1, Mint2, and Mint3. While the amino-terminal region shows little sequence conservation, the carboxy-terminal region is highly conserved and contains a phosphotyrosine-binding (PTB) domain and a PSD-95/DLG-A/ZO-1 (PDZ) domain, both serving as protein interaction modules.
- Mint1 and Mint2: Expressed mainly in the brain; bind to Munc18, a key component of synaptic vesicle fusion.
- Mint3: Ubiquitously expressed, lowest in brain and testis; does not interact with Munc18.
- Functional relevance: Mint3 interacts with the Alzheimer’s disease-related amyloid precursor protein (APP) through PTB and PDZ domains, potentially linking APP to transport machinery and influencing its metabolism.
Abnormal APP metabolism is implicated in early-onset Alzheimer’s disease.
