Target Information

Peroxiredoxin (Prx) is a growing peroxidase family, whose mammalian members are known to be involved in cell proliferation, differentiation, and apoptosis. Many isoforms (about 50 proteins), classified by amino acid sequence homology, contain active site cysteine residues and exhibit thiol-specific antioxidant activity across all kingdoms.

Mammalian Prxs consist of six members grouped into typical 2-Cys, atypical 2-Cys, and 1-Cys Prx. Except for Prx VI (1-Cys Prx subgroup), the other five 2-Cys Prx isotypes have thioredoxin-dependent peroxidase (TPx) activity utilizing thioredoxin, thioredoxin reductase, and NADPH as a reducing system.

Mammalian Prxs are 20–30 kDa in size and vary in subcellular localization:

• Prx I, II, and VI: cytosol
• Prx III: mitochondria
• Prx IV: endoplasmic reticulum and secretory pathways
• Prx V: peroxisome, mitochondria, and cytosol
