Product Specific Information

Specificity: This antibody is specific for murine CD40, a 48 kD type I transmembrane glycoprotein also known as Bp50.

Target Information

The CD40 antigen is a single-chain glycoprotein that belongs to the tumor necrosis factor/nerve growth factor superfamily and shows significant homology to the Hodgkin’s disease-associated antigen, CD30. CD40 is present on all B cells except plasma cells and is also expressed on some epithelial cells, carcinomas, and lymphoid dendritic cells.

CD40 plays a key role in mediating various immune and inflammatory responses, including:

  • T cell-dependent immunoglobulin class switching
  • Memory B cell development
  • Germinal center formation

AT-hook transcription factor AKNA is reported to regulate CD40 expression, facilitating homotypic cell interactions. The adaptor protein TNFR2 interacts with CD40, mediating signal transduction. Interaction between CD40 and its ligand is necessary for amyloid-beta-induced microglial activation, an early event in Alzheimer’s disease pathogenesis. Two alternatively spliced transcript variants encoding distinct isoforms have been reported.

Diseases associated with CD40 dysfunction include:

  • Type 3 Hyper-IgM immunodeficiency
  • CD40 ligand deficiency
