Product Specific Information

The RMST2-2 monoclonal antibody reacts with ST2 (IL-33 receptor, IL-33R), a member of the IL-1 receptor family. ST2 forms a ligand-dependent dimer with IL-1R accessory protein, and the IL-33/ST2 interaction induces TH2 cytokine production.
ST2 has both membrane-bound and soluble isoforms; the soluble form acts as a decoy receptor blocking IL-33/ST2 interactions. It is expressed by TH2 lymphocytes, mast cells, eosinophils, basophils, innate lymphocytes, smooth muscle cells, and endothelial cells.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on mouse bone marrow-derived mast cells at ≤0.5 µg/test (100 µL final volume). Optimal titration recommended.

Super Bright 600 is a tandem dye excited by the violet laser (405 nm) and emits at 600 nm. Use a 610/20 bandpass filter and ensure instrument compatibility.

Using multiple Super Bright dyes: Use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize polymer interactions.
Light sensitivity: Protect vial and samples from light.
Fixation: Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333).

Target Information

IL-33R (ST2) is a receptor for IL-33 and a member of the IL-1 receptor family. It contains three Ig-like extracellular domains interacting with IL-1RAcP and IL-33. IL-33 binding activates NF-κB and MAPK pathways via adaptor proteins (MyD88, IRAK1, IRAK4, TRAF6), leading to cytokine release (IL-5, IL-6, IL-8, IL-13).
Two isoforms exist: ST2L (membrane-bound) and ST2S (soluble). The soluble form acts as a decoy receptor. IL-33R is expressed mainly on mast cells, basophils, eosinophils, and in mouse on ILC2 and Th2 cells. It is implicated in allergy, arthritis, atherosclerosis, and sepsis.
