IL-33R, also known as ST2, is a receptor for IL-33 and a member of the IL-1 receptor family. Its extracellular domain contains three Ig-like domains that interact with co-receptor IL-1RAcP and ligand IL-33. Binding of IL-33 induces complex formation with adaptor proteins (MyD88, IRAK1, IRAK4, TRAF6), activating NF-κB and MAPK pathways, leading to cytokine release (IL-5, IL-6, IL-8, IL-13).
Two main isoforms exist due to alternative splicing: ST2L (membrane-bound) and ST2S (soluble). The soluble form may act as a decoy receptor modulating IL-33 signaling.
IL-33R is primarily expressed in mast cells, basophils, and eosinophils, and in mice, also on ILC2 and Th2 cells. IL-33R signaling is implicated in immune conditions such as allergies, arthritis, atherosclerosis, and sepsis.
