Target Information

TRF1 is a negative regulator of telomere length maintenance. Two proteins, tankyrase and Tin2, which bind to mammalian TRF1, have been identified.
Tankyrase localizes to telomeres by binding TRF1 through its ankyrin repeats and exhibits PARP activity, functioning as acceptors for ADP-ribosylation.
ADP-ribosylation of TRF1 diminishes its ability to bind telomeric DNA, suggesting telomere function is regulated by poly(ADP)-ribosylation.
TIN2 protein interacts with TRF1, localizes to telomeres, and is essential for proper regulation of telomere length.
TIN2 and TRF1 are constitutively expressed and act together to counterbalance telomere elongation by telomerase, though endogenous expression levels are low and may be difficult to visualize by immunostaining.
