Target Information
CD73 (Ecto-5′-nucleotidase, 5′-ribonucleotide phosphohydrolase) catalyzes the conversion of purine 5′ mononucleotides to nucleosides at neutral pH, with AMP as the preferred substrate.
It forms a dimer of two identical 70 kDa subunits attached to the plasma membrane via a glycosyl phosphatidyl inositol linkage.
CD73 serves as a marker of lymphocyte differentiation, and its deficiency is associated with several immunodeficiency disorders.
Other 5′-nucleotidase forms exist in cytoplasm and lysosomes, distinguishable by substrate affinities, magnesium ion requirements, ATP activation, and phosphate inhibition.
The CD73 gene is mapped to chromosome 6q14–q21.
Defects in CD73 can lead to joint and arterial calcification, as well as intestinal tuberculosis.
Two transcript variants encoding different isoforms of CD73 have been identified.
