Product Specific Information

This MR6F3 antibody recognizes mouse CD206, also known as Macrophage Mannose Receptor (MMR) or Mannose Receptor C, Type 1 (MRC1).
CD206 is a 175-kDa type I integral membrane glycoprotein expressed in macrophages, dendritic cells, and endothelial cells. It belongs to the C-type lectin family and mediates endocytic uptake of fungal, bacterial, protozoan, and viral antigens, playing a key role in immune defense and regulation.

CD206 is a marker for M2 macrophages. Its expression is induced by IL-4, IL-13, M-CSF, IL-6, IL-10, and glucocorticoids, while TNF-α, IFN-γ, TGF-β, and LPS down-regulate it.
Only a small fraction of CD206 is found on the cell surface; therefore, intracellular staining is recommended.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Tested by intracellular staining of resident peritoneal macrophages. Recommended ≤ 0.125 µg/test (100 µL final volume). Cell number may range from 10⁵ to 10⁸ per test. Antibody titration is recommended for optimal performance.

Fluorochrome Information:
PerCP-eFluor 710 emits at 710 nm and is excited by the blue laser (488 nm). It can replace PerCP-Cyanine5.5. Recommended filter: 710/50 (alternative: 695/40). Ensure instrument compatibility.

Light Sensitivity: Tandem dye sensitive to photo-induced oxidation. Protect from light.
Fixation: Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-57).

Target Information

CD206 (MSR, Mannose receptor, MRC1) is a 175 kDa transmembrane protein belonging to pattern recognition receptors. It is expressed in macrophages, dendritic cells, endothelial cells, and sperm cells, playing a role in innate and adaptive immunity.
It mediates recognition and uptake of macromolecules such as modified lipoproteins, AGEs, and amyloid β-protein. CD206 is implicated in atherosclerosis and Alzheimer’s disease.
Three isoforms (types 1–3) are generated by alternative splicing; types 1 and 2 are functional, while type 3 has a dominant negative effect by inhibiting endocytosis.
Diseases associated with CD206 dysfunction include leprosy and Gaucher’s disease.
