Target Information
Tryptase gamma (TPSG1), also known as serine protease 31 or transmembrane tryptase, is an enzyme encoded by the TPSG1 gene in humans. Tryptases belong to the trypsin-like serine protease family (peptidase family S1) and are enzymatically active only as heparin-stabilized tetramers, resistant to known endogenous proteinase inhibitors.
Several tryptase genes are clustered on chromosome 16p13.3, including four functional genes: alpha I, beta I, beta II, and gamma I. Each has allelic variants (e.g., alpha II, beta III, gamma II). Beta tryptases are mainly expressed in mast cells, while alpha-tryptases are predominant in basophils. TPSG1 differs from other family members by having a C-terminal hydrophobic domain that may serve as a membrane anchor. Tryptases are implicated as mediators in asthma and other allergic or inflammatory disorders.
