Target Information

The progesterone receptor (PR) is a member of the steroid family of nuclear receptors. It mediates the physiological effects of progesterone, which plays a central role in reproductive events associated with the establishment and maintenance of pregnancy.

PR exists as two forms:

  • Form A (94 kDa)
  • Form B (120 kDa)

These forms arise due to alternative translation initiation sites.
PR-B is transcriptionally active and responsible for activating genes related to endometrium maintenance, pregnancy, and inhibition of ovulation.
PR-A differs by a 165 amino acid deletion at the N-terminus, exposing a 140 amino acid inhibitory domain that represses steroid hormone transcriptional activity.

In its inactive state, PgR forms a multiprotein complex including heat shock proteins and immunophilins. Upon progesterone binding, the receptor undergoes conformational change, dimerizes, and binds to progesterone response elements (PRE), activating transcription.

A null mutation in the PGR gene leads to pleiotropic reproductive abnormalities.
