Target Information
TRIB1, also known as gprotein coupled receptor induced, skip1, gig2, c8fwATP, functions in protein binding, protein kinase inhibitor activity, and protein serine/threonine kinase activity.
Overexpression of TRIB1 in HeLa cells represses IL8 promoter basal activity by inhibiting AP1 activity. It also inhibits oncogenic Ras-driven and MEKK1-mediated AP1 activation, while enhancing ERK activation.
Coimmunoprecipitation and yeast 2-hybrid assays show that MEK1 interacts with TRIB1 and TRIB3, and MKK4 interacts specifically with TRIB1. Cotransfection of MKK4 enhances TRIB1 levels, indicating that TRIB-MAPKK interaction stabilizes TRIB1.
