Target Information
Sonic Hedgehog (SHH)는 배아 발생 중에만 발현되며, 초기 배아의 패턴 형성에 중요한 역할을 합니다. SHH는 복측 신경관, 사지의 전후 축, 복측 체절의 형성에 관여하는 주요 유도 신호로 알려져 있습니다.
이 단백질은 전구체 형태로 합성되어 자가 촉매적으로 절단되며, N-말단 부분은 용해성으로 신호 전달 기능을 가지며, C-말단 부분은 전구체 처리에 관여합니다. C-말단 산물은 N-말단 산물에 콜레스테롤 잔기를 공유 결합시켜 세포 표면에 제한함으로써 배아 내 자유 확산을 방지합니다.
이 단백질 또는 신호 전달 경로의 결함은 전뇌가 좌우로 적절히 분리되지 않는 기형인 holoprosencephaly (HPE)를 유발할 수 있으며, 얼굴 기형으로 나타날 수 있습니다. 또한, SHH 관련 변이는 VACTERL 증후군(척추, 항문, 식도, 신장, 심장 및 사지 기형)을 유발할 수 있는 것으로 알려져 있습니다.
