Product Specific Information
Description:
The 17A2 monoclonal antibody reacts with the mouse CD3 complex. CD3 subunits (gamma, delta, epsilon) are essential for proper assembly and surface expression of the TCR complex. CD3 is expressed by thymocytes and all mature T cells. Binding of 17A2 to CD3 initiates intracellular signaling leading to activation and proliferation.
Applications Reported:
Reported for flow cytometric analysis and functional activity (in vitro stimulation).
Applications Tested:
Tested by flow cytometric analysis of mouse thymocytes and splenocytes. Recommended ≤1 µg/test (in 100 µL volume). Optimize empirically for cell number (10⁵–10⁸ cells/test).
Fluorochrome Information:
Super Bright 780 is excited by a 405 nm violet laser and emits at 780 nm. Recommended 780/60 bandpass filter. Protect from light due to photo-induced oxidation sensitivity.
Compensation Notes:
Higher compensation values may occur in violet 450/50 channel when using UltraComp eBeads or AbC Total Antibody Compensation beads. In such cases, set compensation with stained cells.
Staining Buffer Recommendation:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific interactions.
Fixation:
Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer or 1-step Fix/Lyse Solution with minimal loss of brightness or FRET efficiency.

Target Information
The CD3 subunit complex transduces antigen-recognition signals into T cells and regulates TCR expression. Subunits (gamma, delta, epsilon, zeta) are part of the immunoglobulin superfamily encoded on chromosome 11. CD3 is expressed on 68–82% of normal peripheral blood lymphocytes and 65–85% of thymocytes, but not on B or NK cells. CD3 defects are associated with immunodeficiency and altered T-cell populations in autoimmune diseases.
