Target Information

IkB-alpha is a 40 kDa protein that functions to inhibit NF-kappaB activity through protein-protein interactions with NF-kappaB dimers in the cytosol. This interaction masks the nuclear localization sequence of NF-kappaB, preventing its translocation to the nucleus. Various stimuli, including TNF-alpha, IL-1 beta, chemokines, PMA, growth factors, LPS, UV irradiation, viral infection, and other stresses, can activate NF-kappaB by inducing degradation of IkB-alpha.

In humans, the IkB-alpha gene is located on chromosome 14q. Activation of NF-kappaB requires phosphorylation of IkB on specific serine residues (Ser32 and Ser36), leading to its degradation. IkB kinase alpha (IKKα) interacts with and phosphorylates IkB-alpha at these sites. IKKα, in complex with IKKβ and IKKγ, is essential for NF-kappaB activation in response to proinflammatory cytokines, regulated by NF-kappaB inducing kinase (NIK).
