Target Information

Mitochondrial transcription factor A (TFAM) is a key nuclear-encoded protein that translocates to mitochondria, where it maintains and regulates mitochondrial DNA (mtDNA) replication and transcription. TFAM contains an LC3 interacting region (LIR) motif, enabling interaction with autophagy proteins like LC3, which is critical for mtDNA quality control and inflammation regulation. TFAM levels influence mitochondrial function across tissues—high expression can suppress mtDNA transcription in skeletal muscle, while in cardiac tissue, balanced TFAM-to-mtDNA ratios support oxidative phosphorylation (OXPHOS). Dysregulation of TFAM is linked to mitochondrial dysfunction, neurodegeneration, and diseases associated with mtDNA stress.
