Product Specific Information

The A7R34 monoclonal antibody reacts with mouse CD127, the high-affinity alpha subunit of the mouse IL-7 receptor. CD127 is expressed by immature B cells in bone marrow, double-negative (CD4⁻CD8⁻), single-positive (CD4⁺ and CD8⁺), but not double-positive thymocytes. Mature peripheral T cells express CD127 at low levels.
A7R34 inhibits IL-7 binding to its receptor and is used in vivo and in vitro to study IL-7 roles in T/B cell development and activation. It blocks the binding of SB/199, another antibody recognizing mouse CD127.

Applications Reported: Flow cytometric analysis.
Applications Tested: Flow cytometric analysis of mouse thymocytes and splenocytes at ≤0.5 µg/test (100 µL final volume). Optimize antibody titration for best performance.

Target Information

CD127 (Interleukin-7 receptor α, IL7R) is a glycoprotein regulating lymphopoiesis. The active receptor is an α/γ chain heterodimer. The γ(c) chain activates signal transduction, while the α chain determines specific signaling via cytoplasmic molecules.
CD127 promotes proliferation of precursor B cells, thymocytes, T cell progenitors, and mature CD4⁺/CD8⁺ T cells. Dysfunction is associated with severe combined immunodeficiency (SCID) and related T⁻/B⁻/NK⁺ SCID forms.
