Product Specific Information

This antibody detects endogenous levels of HER4 only when phosphorylated at Tyr1242.

Target Information

Oncogene ERBB4 (HER4) is a member of the type I receptor protein tyrosine kinase subfamily that includes EGFR, ERBB2, and ERBB3. ERBB4 binds and is activated by neuregulins and other factors, inducing various cellular responses such as mitogenesis and differentiation.

ErbB4 is activated by neuregulins and NTAK (neural and thymus-derived activator for ErbB kinases). It serves as a receptor for heregulin and mediates HGL-stimulated tyrosine phosphorylation of itself. Like other protein kinases, ErbB4 regulates cellular metabolism, transcription, cell cycle progression, cytoskeletal rearrangement, cell movement, apoptosis, and differentiation.

ErbB4 is predominantly expressed in several breast carcinoma cell lines and in normal skeletal muscle, heart, pituitary, brain, and cerebellum. High expression is observed in T47-D, MDA-MB-453, BT-474, and H3396 cell lines, while intermediate levels appear in MCF-7, MDA-MB-330, and MDA-MB-361. Expression is low or absent in MDA-MB-231, MDA-MB-157, MDA-MB-468, and SKBR-3. Elevated ErbB4 levels in certain tumor cell lines suggest a possible role in malignancy.
